Will email verification slow down checkout?
No. Verification typically answers in under half a second, or 1 to 3 seconds when a live SMTP check is needed. The Magento module fails open by default, so a network problem never blocks a sale, and the WordPress plugin caches each result for 10 minutes so the live check and the order use a single credit.

Will it block real customers?
You choose the rules. Most stores block disposable and undeliverable addresses and let everything else through.
